目的

探讨颐脑解郁复方对卒中后抑郁(post-stokedepression,PSD)大鼠受损脑组织的神经保护机制。
方法

将SD大鼠分为正常组、假手术组、卒中组、PSD组、PSD西药组、PSD中药组6组。采用大脑中动脉梗塞(middle cerebral artery occlusion,MCAO)所致的局灶性脑缺血模型和局灶性脑缺血复合孤养、束缚应激的PSD大鼠模型,从造模开始到实验结束,各组每日灌胃1次。第2,4,6周应用Western Blot法观察海马TLR2,TLR4和NF-κB蛋白的表达。灌胃结束后用HE染色方法观察海马CA3区的病理变化。
结果

4周时各组大鼠海马TLR2,TLR4蛋白表达差异有统计学意义(

0.01),PSD组TLR2,TLR4表达均明显高于正常组、假手术组、卒中组、PSD西药组、PSD中药组(

0.01)。第2,4,6周时各组大鼠海马NF-κB蛋白表达差异均有统计学意义(

0.01),PSD组NF-κB表达高于正常组、假手术组、PSD西药组、PSD中药组(

0.01)。与正常组相比,PSD组细胞形态不规则,结构不完整,染色较深,排列稀疏、紊乱,细胞层数少,细胞数量明显减少。与PSD组相比,PSD西药组及PSD中药组大鼠海马CA3区细胞形态较规则,结构完整清晰,排列相对整齐,细胞层数增多,数量增加。
结论

颐脑解郁复方治疗PSD的可能机制是通过减少TLR2,TLR4和NF-κB蛋白的表达,从而减轻免疫炎症反应,修复海马区神经细胞。
Objective

To explore the neuroprotective mechanism of Yinao Jieyu Compound on damaged brain tissues in rats with post-stroke depression(PSD).
Methods

SD rats were assigned to the normal control group
sham operation group
stroke group
PSD group
Western medicine group and traditional Chinese medicine group.The focal cerebral ischemia model induced by MCAO and the PSD rat model of focal cerebral ischemia combined with orphan and restraint stress were used.From the beginning of the model to the end of the experiment
each group was intragastrically administered once a day.The expressions of TLR2
TLR4 and NF-κB protein in hippocampus were observed by the Western Blot method at the 2nd
4th and 6th weeks.After gavage
HE staining was used to observe the pathological changes in the hippocampal CA3 area.

At the 4th week
the differences in the expressions of TLR2 and TLR4 proteins in the hippocampus of each group were statistically significant(

Conclusions

Yinao Jieyu Compound may treat PSD by reducing the expressions of TLR2
TLR4 and NF-κB protein to reduce the immune inflammatory response and repair nerve cells in hippocampus.
